Abstract:ObjectiveTo establish an RPHPLC method for the determination of Hyperin in Manchurian rhododendron Leaf.MethodsThe DiamonsilTM C18 (4.6 mm×250 mm,5 μm) was used. The mobile phase was methanol-5%phosphoric-triethylamine(44∶56∶0.37).The flow rate was 1.0ml/min,the detective wavelength at 360nm.20μl sampler and external method were used. Results A satisfactory separation was obtained.The calibration curve was linear over the range of 0.05~0.28μg for ribavirin(r=0.999 8,n=6).The average recovery of ribavirin was 99.83%(n=5)with RSD of 0.90%.ConclusionThe method is quick and simple with good reproducibility. It can be used for the quality control of Hyperin in Manchurian rhododendron Leaf.
黄酮苷类成分在用HPLC测定时易拖尾,流动相中加入三乙胺可抑制其拖尾[3]。曾使用了低浓度(0.05%,0.1%,0.5%)的醋酸溶液和0.05%磷酸溶液,峰形对称性不好,后用甲醇-0.5%磷酸溶液(44∶56)得到较好的效果。但实验中发现仍有轻微的拖尾现象,经反复实验发现在1 L上述流动相中加入3.7 ml
